Determination of Maize Dwarf Mosaic Virus (MDMV) in Seeds of Some Maize Breeding Lines and Virus Elimination by Thermoterapy Treatments

- This study was carried out in inbred line maize trial fields of Sakarya Agricultural Research Institute in 2008 and 2009 years. Eighteen leaf samples of 10 inbredmaize lines were collected for detection of Maize dwarf mosaic virus (MDMV). It was detected in 5 out of 10 inbredmaize lines by DAS-ELISA. These results were confirmed by RT-PCR. Seeds of infected 5 inbred maize lines belong to 2006, 2007 and 2008 years were tested by DAS-ELISA for determination of infection source. According to the results of seeds showed that only one inbredmaize lines was found to be infected with MDMV. Seeds of infected inbredmaize line were dissected to their parts (endosperm and embryo) and was detected MDMV in the embryos by DAS-ELISA. Infected seeds were treated to dry heat treatments for elimination of virus at 40˚C, 50˚C and 65˚C temperatures for 48 hours. The titerof MDMV decreased significantly, especially at 50 ˚C and 65 ˚C, but virus was not completely eliminated. Keywords : MDMV, maize, RT-PCR,thermotherapy
